According to "Maternal-Fetal Toxicology" by Koren, "most of the antihistamine compounds available over-the-counter have not been implicated in fetal or pregnancy problems."

A large study of exposure in pregnancy showed that only with diphenhydramine (Benadryl) was there a significant association with fetal defect. This medication was thought to cause an increased incidence of cleft lip and palate but recent studies have not been able to prove this. "The use of antihistamines in the first trimester does not appear to increase the risk for major birth defects."
